We are using a trichoderma asperellum strain as biocontrol agent, I would like to know some techniques to maintain its biocontrol properties. Does somebody have tried cultivating it on soil to maintain its growing characteristics? Dual culture? Which parameters do you determine?
It is not uncommon that repeated subculture under laboratory conditions can cause the degeneration of pathogenic wild phenotype reactions. For you biocontrol Trichoderma as a hyperparasite I would suggest maintaining the fungal culture on the host fungus of interest. The fungal host for the hyperparasite should be kept on minimal nutrient media as rich media stimulates quicker degeneration from the original wild pathogenic type. As you suggest the employing of dual cultures would allow you to select for the persistence of antibiotic zones of inhibition which can be important factor expression for biological control. If you are using Pythium as a target pathogen for control this would be the source for growing of the hyperparasite. The use selective oomycete fungicide would allow the hyperparasite to increased and preserved and the unwanted host could be eliminated without eliminating the hyperparasite by using the oomycete selective fungicide as a plating treatment after the increase but before the application of biocontrol.
