I did an ELISA to find salivary Oxytocin concentration in adolescents. Although the subjects were able to give different volumes of saliva (200-1000 μl) the variance in concentrations found in the test is small. I should mention that all extracted Oxytocin samples were diluted in the same volume of assay buffer for the ELISA so I was expecting to see a smaller concentration of Oxytocin in samples with smaller amounts of saliva, but actually the concentrations were similar.

Does that make sense? Is it possible that the volume of saliva is not correlated with amount of Oxytocin in it?
