I am wondering whether reliability of any developed questionnaire depends on population, especially their age. My sample is stratified into rural urban; and higher and lower socio economic groups. Does this affects reliability of the questionnaire? The reliability of pilot questionnaire without stratified sample was good but the final questionnaire with bigger sample ended up with cronbach's alpha of 0.642. What would be the reason? The age of my participants was between 12-15 years.

It depends on the questionnaire and what age group it was validated for use with. Some questionnaires may seem to be applicable to young people, but have only been validated with adults, and so would not be appropriate. Look at the research identifying the original questionnaire to see. Problems can occur, especially with Likert-type formats, which carry some negatively-phrased questions, as they can be confusing (young people may not be able to identify which end of the scale they want to select; they can also select an answer whatever their actual opinion, without any facility to check whether this reflects what they actually think! I've written a paper on this for Sage's online research methods platform. It's much more complex to use these tools with young people than you might think!
The age of informants could have a material bearing on the responses to questions. This obviously has implications for reliability
