I would like to monitor hedgehog signaling using good monoclonal antibodies against the Gli isoforms in mesenchymal muscle cells isolated from C57BL6/j mice.
All other suggestion for monitor hedgehog signaling are well accepted.
There is no Good commercial antibodies for the Glis to my knowledge. But several lab made polyclonals have been published at least for Gli2 and Gli3. You could try to request some from them. But If you're trying to asses the activation of the pathway, I would not waste precious antibodies but I would recommend doing qPCR on Gli1 which is upregulated upon activation of the pathway. Primers are published, you can take a look at the 2012 and 2014 dev cell papers on IFT25 and 27 from our lab (Pazour Lab).
Currently, I didn't find a good antibody against GLIs. However I decided to overcome this problem using qPCR. There are several good primer at this purpose.
