I want to do IHC for slides for a membrane bound marker after the FFPE cells have been cytospun onto a slide. I'll be using the positively stained cells to do further nuclear analysis so the membrane can't be destroyed, and has to be associated with the nucleus and not shattered all over the place. Does cytospinning destroy or shatter cell membranes?

I completely agree with Sebastian. We use cytospinning frequently to concentrate diagnostic samples from fluids.

Cytospin alothough is know to cause nuclear as well as cytoplasmic distortion. But as suggested by Sebastian maintaining the acceleration would definitely be of help. However membrane filter technique is another option which will provide even better results.
