For instance, there is no growth when the initial concentration of bacteria in a medium with high ionic strength is adjusted to 10^6 cell/ml, but the growth happens when it is adjusted to 10^8 in the same medium.
how do you interpret this?
---------------------------------------------updated description:
- The bacteria is halophilic.
- I mean the cell concentration in solution at T=0 is adjusted to 10^6 or 10^8 cell/ml.
- Assuming the destination solution is mixture of solution A and B. Salinity of the destination solution is ~ 130 mS/cm. in order to adjust the bacteria concentration to 10^6, 10^7 and 10^8 cell/ml in this solution at T=0, I followed the following steps:
for 10^8: I took 1ml of origin bacterial solution (10^9 cell/ml, salinity of the fresh solution without bacterial was ~ 25 mS/cm). it was centrifuged and the supernatant was replaced with solution A (salinity= ~65 mS/cm). The destination solution (9ml) was then inoculated with the whole 1ml solution.
for 10^7: the same as above but the destination solution (9.9 ml) was inoculated with 0.1ml of that 1ml centrifuged solution.
for 10^6: I took 0.1ml of origin bacterial solution (10^9 cell/ml, salinity of the fresh solution without bacterial was ~ 25 mS/cm). then it was diluted with 0.9 ml of solution A (salinity = ~ 50 mS/cm). it was centrifuged and the supernatant was replaced with solution A (salinity= ~65 mS/cm). The destination solution (9.9 ml) was then inoculated with 0.1ml of the 1ml centrifuged solution.
The proportion of A and B in destination solutions for all concentrations is the same.
The bacteria were taken from the same origin bacterial solution.
THERE IS NO GROWTH FOR 10^6, SMALL GROWTH FOR 10^7, HIGH GROWTH FOR 10^8! WHY!?
