I'm designing an experiment that will utilize CRISPRi in a diatom. I know that the main difference between regular CRIPSR and CRISPRi is that the Cas9 protein is deactived enzymatically. I also know that there are numerous Cas9 proteins which are recommended for different species. My question: does the choice of Cas9 protein matter for CRISPRi, given that the enzyme will be deactivated anyways?
