In Platinum-E cells , by co-transfecting pBabe retroviral transfer vector and pVSVG vector, can retrovirus be generated. We have been successful with mscv retroviral backbone plus VSV-G vector, but not sure how about pBabe backbone ?
pBabe backbone will work. Different retrovirus vectors shouldn't matter. The packaging cell line is traditionally to be 293 or its derivatives because you can achieve high transfection efficiency and gene expression level. For packaging, you just need a retrovirus vector together with Gag, Pol and Env (envelope) genes.
Platinum-E is specialized to make retrovirus ecotropic (infecting mouse and rat only). It has the envelope protein stably expressed. If you use VSV-G, then your virus should be pantropic (can infect many other species). It's not good idea to co-transfect VSV-G in Platinum-E cells if you want to use your virus to infect human cells.
