Do you think that MMP-14 antibodies could be injected intraarticular and still serve the purpose of blocking this proteinase? Would this not cause immunological response?
Thank you John, although all proposed compounds are not selective for MMP-14. They bond to broad spectrum of other MMPs... As I will use this compounds in vivo I need them to be selective, otherwise I could not claim the effect through MMP-14
you may have trouble with that. I read that Britsh Biotechnology spent hundreds of millions of dollars, unsuccessfully, trying to find selective MMPx inhibitiors. The problem is that the binding sites are so similar, so the ligand would need to be very big. So there is a conflict between the molecule's size and the selectivity.
Thank you for your help! I don't know much about the topic yet. The only "claimed to be" selective inhibitor of MMP-14 seems to be DX-2400 from Dyax. I've contacted the company and waiting for replay.
I hope for us all that MMPs pharmacology will jump forward soon :)
