I am looking to conduct transport studies in CaCo-2 cells using three fluorescent dextrans namely, Fluorescein Salt (MW~3.7KD), FD4 (MW~ 3-5KD) and FD20S (MW~20KD). All three of them are from Sigma-Aldrich. Though, generally they are used in a 1mg/ml concentration, but, I am still wondering is there any other variation to it which someone has used before?
We use the 1 mg/ml concentration, that works really nice. Just out of curiosity, why do you want to vary the conc, costs or another issue?
Thank you Birger. Actually, I did read a few papers and most of them used the same concentration. We are doing the experiment for the first time, so, we are conscious about the toxicity of the tracer to the cells. That's why, I wanted to know if there are any variations to the concentration.
ah, I see. We haven't experienced cytotox but I must admit , we haven't been looking either..But if you monitor TEER after the exp. (and compare to a control because TEER will always drop over time) you will know about monolayer integrity, which is often the first indicator of tox in the Caco's.
Thanks Birger. We did the same. The concentration was not toxic to the cells. So, we will be using the same for further experiments.
