Classical site-directed mutagenesis polymerases don´t displace the mutagenic primers and therefore introduce changes. I have had VERY good experiences in mutagenesis with the KOD Hot Start Polymerase (Merck) before and am curious about this Phusion Flash High-Fidelity PCR master mix by Thermo Fisher.
