I am trying to isolate stem cells form the subventricular zone (SVZ) or subgranular zone (SGZ) in rat with very bad results. Thanks a lot for your help.
Can you elaborate more on your problem? It is hard to give advice without knowing the problem.
I have generated primary rat stem cell cultures. What are your problems? What's your protocol? Happy to answer specific questions.

I have been isolating rat stem cells for years, but now the main problem is I cannot get neurospheres on culture derived from stem cells. When finishing my isolation protocol, after 3-4 days on culture I just have a cellular monolayer adhered on plate instead of floating stem cell-riched spheres. I have checked the process a thousand times and I am completely lost. Even I use non-treated multi-well plates.
Any suggestion?

I only cultured the stem cell from mouse SVZ, but I think the procedure will be similar.I advice you see the video on JOVE "isolation and culture of mouse neural precursors ", Hope it will help
