I wish to investigate the effect of (recombinant) MPO on in vitro cultured murine primary cells, such as T cells. MPO is mostly known to affect cells through its enzymatic activity, but has also been shown to exhibit cytokine-like properties through binding of cell surface proteins.

In order to examine the effect of its enzymatic activity, H2O2 and 4-ABAH (specific MPO inhibitor) will be added to the culture.

Both recombinant mouse and human MPO are available from R&D systems. However, I was told that up until recently recombinant mouse MPO was not available and therefore, recombinant human MPO was used e.g. as reference in MPO activity assays. I have performed one MPO activity, too, where rmMPO did not at all work as a standard reference, so I used rhMPO instead, which worked fine. Hence, I am questioning whether rmMPO is "trustworthy" in a way.

Does anyone have experience with these reagents and this experimental set up? I am mainly wondering whether I can/should use rhMPO instead of rmMPO with murine primary cells.
