I am growing the colorectal carcinoma cell line LoVo in DMEM with 10% FBS and 1% P/S, and I have encountered the problem of waste accumulation in the medium mentioned above, and it seems like the debris makes cells stop proliferating after around 2–3 days after passaging, even though there is still much space to grow, and the nutrients shouldn't be depleted. Washing away the debris and changing the medium to a fresh one does help. But is there a way to avoid this and help them grow happily while there is still space and sufficient nutrients? I have tried RPMI medium instead of DMEM, it doesn't make a difference, also have tried releasing them with accutase instead of trypsin, and also tried growing them at different densities. None of that solved the problem.
