The calculation will depend on the assay method. You will need to provide the entire assay method, as there are different types of assays one can use.
If you are using NADH or NADPH for the assay, you can determine aldose reductase activity by evaluating the decrease in OD at 340 nm.
Hello, I know this conversation is a bite old but I think it is the right place to ask my questions. I have expressed and purified recombinant human aldose reductase. I have no problem recording its activity using NADPH, however, I cannot follow its activity using NADH using the decrease at 340 nm. Does the enzyme work the same way with NADH? Do I need to change my reaction conditions? Is there litterature on Aldose reductase activity with NADH?
