I'm after something that will allow me to measure the stomatal guard cells responses to certain stimuli, so the media used for the peel has to be rather flexible. I have thought about a fast drying medical silicone adhesive, but am not sure where to find this in Australia
I am not familiar with grapevine leaves but with plant anatomy. If there are many trichome on lower epidermis of grapevine leave it is a problem to remove cuticle with stomata alone. There are some technique for plant with many trichome on leaves. The technique which remove the cuticle and stomata layer from leave, I never done before. But if you need this technique I need to find from some of my book after.
One technique that I use with successful for removing stomata from stomatal cryp of Casuarina (brenchlet) leave is.. boil the leave in 2% NaOH ,after the leave are soft, then you remove the trichome or try to find the small specimen. Use needdle and small brush to remove mesophill until you got the stomata. These processes you need to do under steriomicroscope. Hope you can find the solution.
