I am trying to analyse if my complex interacts with DNA by UV-spectroscopy but I don´t have any experience in these type of experiments, so if someone can send me a reliable protocole it would be fantastic. I already have calf-thymus DNA and Tris-buffer.
You can obtain the UV spectra of bare DNA and DNA+small concentrations of your comples and see if you have a hyperchromic (damage of the secondary DNA structure) or hypochromic effect (stabilization of DNA via electrostatic interactions or intercalation). Usually the interaction could be detected by:
- a new peak is appeared
- the original peak is shifted (red or blue)
try first with DNA spectra and then add the complex step wise and see what happen
