Hello everybody, i am looking for a protocol to quantify sjTREC and ß-TREC by PCR on PBMC. I read some papers to find one, but it seems that there is not a clear defined protocol for it. Do you have any recommandations ?

In fact, my main challenge is creating the positive control plasmid for generating the standard curve in qPCR. I've reviewed several protocols, but most of them are quite labor-intensive and involve multiple cloning and amplification steps. I'm looking for a simpler and more efficient approach—do you have any suggestions?
