I need to the Hyp assay on mouse bones. Most of the literature describes the assay performed on lung or skin tissues. One of the few protocols mention to start with dry bone but how to proceed from that to hydrolysis step is not mentioned. How does one normalize the weights when different groups are considered? Does one need to decalcify the bone to measure Hyp content?
