I am doing flavivirus infection in diploid cell line. After infection, no CPE are observe till 7 days. In the same time titer is also low(103-104 ccid50/ml). Is this cell line is not suitable for flavivirus propagation? or some other factors(like pH, temp, media components) play a role in it?
If virus can propagated for 3-4 times in same cell lines, is there will be chance to increase titer?

Dengue does better in Vero than diploids. Might want to try BHK, PDK, or similar lines.

Thanks for your reply. I was tried in two different cell lines( Vero and FRhL-2). In Vero cells, i got virus titer of 107 pfu/mL but in FRhL-2 cells, the virus titer was not more than 104 pfu/mL.
During plaque assay, the morphology of plaques was also different. Virus supernatent from Vero cells had clear,big plaques while in FRhL-2 cells, plaques size was small.
