I have a drug which interacts with protein. If i perform FTIR at 2 cm-1, i got shift in peak of drug by 2 cm-1 in presence of protein. My question is at what resolution we should perform FTIR experiment for liquid sample, so as to interpret correctly about interaction. Is it possible that if i perform experiment at resolution of 4 cm-1 and there is spectral shift of 2 cm-1, i can interpret it as significant shift. What is role of resolution?
Dear Virender,
In general resolution is the ability to separate two consecutive bands. You can imagine that a 4cm-1 resolution is not enough to characterise precisely a small shift like 2 cm-1. I think you probably need even a lower spectral resolution i.e. 1 cm-1.

Thanks Sir for reply. But is it reasonable to use 1 cm-1 for solid or liquid samples. I have read that that should be applicable to only gaseous samples. I have not seen papers where someone used that high resolution for liquid sample.
Indeed when you analyse just one sample in liquid form, a 4 cm-1 resolution is enough compared with the bandwidth observed in Mid-infrared range. However, if you need to characterise a small shift from 2 spectra, a 1 cm-1 resolution seems to be necessary.
