You mean for a cell sorter? And what tissue? Spleen?
You probably don't need a sorter, unless you are looking for a minor sub population of some sort.
Monocytes... easiest is probably CD14. If you use a sorter, then you can use CD11b+GR1-.
Macrophages...I've never done this... but probably F4/80?
I don´t intend to do cell sorter at this point... I meant fresh isolate cells from blood, spleen and gut. I was wondering if Ly-6C could be a good choice for both monocytes and macrophages
I guess you can use that... But are you going to couple the antibodies to the beads yourself? I didn't know they make isoaltion kits with that marker.
Otherwise, you are probably better off using CD14 for both moncytes and macrophages. Yes, you will have contiminated B cells, but depending on what you do later, it might not matter or it can be easily removed. Or you can probably use a monocyte isolation kit for macrophages too..
