Recently I have extracted RNA from Vero cell line. Unfortunately, the result of agarose gel electrophoresis showed that there is an extra band just right above the 28S rRNA. I have treated the samples with DNase but the band is still exist. I have done PCR to check whether the samples contain any DNA and it showed no DNA contamination. I also sent my samples to bioanalyzer and the result did not show DNA contamination.
Yes, the faint band which sometimes appear above the 28S band contains theY preprocessed rRNA or mRNA called the hnRNA. Usually if their is a gDNA contamination, the band appear just below the wells due to its high molecular weight. Bit if your band has travelled a few centimeters from the well, then it is the preprocessed RNA. So in such case, no need to worry about the DNA contamination..
Thank you Mr. Vishal for the reply. Do you know that is there any paper mentioned about this hnRNA band that appears above the 28S rrna? Because I would like to justify it during my presentation later.
