Hi Adil, we use the Developmental Studies Hybridoma Bank (Iowa University) mouse monocolonal anti-Nestin Rat-401c (the concentrated one, the other one does not work). It works very well 1:200 on 25 um free floating sections, using 1% DMSO and 1% triton in the blocking (Ab diluted in the blocking), O/N at 4°C. It nicely stains the stem cells in the subventricular zone of the lateral ventricle.

Nestin is a marker for stem cells not for differentiated neurons. We use Mouse anti-Nestin (Chemicon; MAB 353) antibody for labeling stem cells.

Thanks Patro, I used the Abcam antibody (mouse IgG), product number ab6142, on adult mouse brain sections. I got almost smiler results to the one you presented in your paper with P21 Hippocampus. I was hoping for better expression, so am going to retry it with more fresh mouse brains.
