For total SOD assay by Kono, (1978) 3.0 ml reaction mixture contained 50 mM sodium carbonate (pH 10.2), 24 µM NBT, 0.1 mM EDTA, 1 mM hydroxylamine, hydrochloride, 0.03% (v/v) Triton X-100 and 70 µl enzyme extract.
Does any one explain me the volume of these components how to make the 3.0 ml reaction mixture?
Do you have any stock solutions of these compounds? What are their concentrations in these stock solutions?

To 1 L of deionized water, add 5.3 g sodium carbonate, 19.6 mg NBT, 29.2 mg EDTA, 69.5 mg hydroxylamine hydrochloride, 300 mg Triton X-100. From this mixture, take 2.93 ml and pour in a tube. Add 70 microliter of sample to the tube. Follow the rest of the assay method (incubation time, wavelenght ...)
