Dear flow cytometry lovers and users,
I am a Fortessa (BD) user since 2010. Now, as a Scientific Researcher at Instituto Adolfo Lutz, we get a grant to buy a flow cytometer. We chose the Cytoflex S (Beckman) and now we are in a doubt. Sellers told us that FACs Melody has the same capacity to analyze cells than an analyzer. At the moment we are just conducting projects in flow cytometry immunophenotyping field. We do not think about sorting cells until now. What do you think about? In your experiences, has a sorter the same capacity as an analyzer flow cytometer to analyze cells (13 colors) ?

In general, flow cytometers are of two types as you have mentioned-
1. Sorter
2. Analyzer
Usually, the dedicated sorters aren't that good for analyzing multicolour immunophenotyping. However, some instruments are of mixed type such as FACS ARIA II or FACS ARIAFUSION can do the type of experiment that you are asking. It is a 15 parameter instrument of which 13 is fluorescent marker.

if the lasers and the filters on the instruments are the same, a sorter and an analyzer should be equally good to perform multicolor phenotyping. However, if you don't intend to sort cells in the near future, I would go for an analyzer with a plate loader, especially if you have many samples. In sorters, you can only feed one tube at the time to the instrument.

I thanks you in advance for your replies. We agree with you and for us, now, the analyzer is better than a dual equipment that does flow cytometry and sorter!

It depends on what you need to analyze which will affect your choice of either sorter or analyzer
Thanks Emmanuel, for now we are not thinking about sorting but as research goes on and the results come maybe we need that in near future!
