Before putting the tissue into the OCT compound, we know that it needs to be fixed and dehydrated through a pretreatment process.

My method is to store fat tissue, liver tissue and skin in 4% paraformaldehyde for 24 hours at 4 ° C.

After soaking, rinse for 2-3 times with PBS and dehydrate in 10%, 20%, 30% sucrose solution until it sinks.

As far as I know, it takes 24 hours for the tissue to settle down in sucrose, but the tissue I have treated does not sink.

Of course, the fat tissue does not sink, and the liver tissue sinks as soon as you insert it. But the skin does not sink.

What is the reason ??
