When transforming DH5α cells with a low-copy number plasmid (pBAD33::chloramphenicol resistance, in my case), I notice no growth in my chloramphenicol plates. I have retried transformation a few times now, remaking my stocks/plates to no luck. I'm wondering if it's possible that a low-copy number plasmid would require longer recovery post-transformation to express sufficient antibiotic resistance gene (manufacturer recommends 1h/37°C/250rpm, I'm thinking closer to 2h). Thoughts?
