Hi everyone, I need to storage Clostridium botulinum strains, and I would like to know if the better option for this is to use Tarozzi media, supplemented with starch. Do you have alternative culture media?
I have never cultured this strain but I have cultured two other Clostridium sp. strains. What I use as a reference point is the DSMZ culture collection website as these generally contain fairly detailed instructions on how to culture, preserve, and revive strains. With regards to the sporulation aspect of your work, are you looking to induce sporulation and then revive strains from their sporulated state? If so, I am not sure how important the medium composition is, although I would make sure to supply cells with ample calcium as, if I am not mistaken, this is a crucial nutrient involved in developing spores. To induce sporulation, many labs pasteurize/use heat at ~80 C which seems sufficient for revival afterwards. In any case, when reviving the spores make sure you're using freshly prepared medium and you allow enough time for the spores to germinate. You can always try checking under a microscope with a negative stain to see if this approach works.
