There are several protocols in the literature described for sequencing the terminal parts of the proteins (most frequently after being digested in 3M HCl for a few minutes in a common microwave oven). However, only some proteins seem to undergo this reaction; the yields, reproducibility and specificity (cutting from the protein ends) are not convincing. Do you have any idea how to improve the reliability and universality of the method?
