I was doing an experiment with Calcipotriol on dermatitis treatment two cell lines MSC and L929 and when I made the first trial with the 96 well plate and I applied 5000 cells per well and applied 10 ^-5 calcipotriol, I observed all cells died but when I applied the same concentration of calcipotriol on 10000 cells per well the cells live in L929 but MSC cell line no, so what's happened?

Searching the Drug bank web site regarding calcipotriol revealed that it is a synthetic derivative of Vitamin D.
the mechanism of action ..
known to play a role in psoriasis and are believed to undergo modulation of gene expression with binding of calcipotriol to the VDR. This modulation is thought to affect gene products related to cell differentiation and proliferation.
As this drug affects the cell proliferation and differentiation , you have to use with causion and use a minimal dose.
So do the experiment again using different minimal doses till you get the suitable one for your work.

I used original calcitriol (1,25 (OH)2 vit.D3 on human melanoma (BLM) cell line. It showed a dose-dependent decrease in cell growth. Since, calcipotriol is a derivative of VitD3, it has the same effect. So you have to titrate the concentration.
