In most Flow Cytometry (FCM) graphs in journal articles of polyploid tissue, they show evidence of polyploidy as multiple peaked histograms.
Does leaf tissue of a polyploid have 2n, 3n, 4n, and 6n cells, or are they all 3n? What are the multiple peaks, different phases of the polyploid cell cycle (G0, G1, S, G2)?

I cannot answer all your questions. But I performed FCM in Tetra-, hexa and octoploids, on mature leafes and got for each one specific peak. Have a look at the attached paper page 4, 1 = tetra-, 2 = hexa-, and 3 = octoploid.
It is very important to use also a diploid control if possible.
3n (triploids) are very unlikely, normally polyploids have an even number since they are otherwise normally not fertile. To my knowledge only e.g. bananas and melons (without seed because of infertility due to unbalanced gametes) should be triploid.
