Semi-landmarks are digitized the same as landmarks, but they are analysed differently. So pretty much you can digitize them the way you prefer.

For example, i used to digitize semi-landmarks on TPSDig using a drawing function and latter converted the data into landmarks (there are tools to do all of those in the software). I could have digitized them directly with the landmark function, but i wanted equally spaced landmarks.

Hi Epa, you can use the draw background curve tool in tps.Dig (yellow pencil) to draw the curves and re-sampled the curve with a number of point which can append to landmarks using tps.util.

You can quantify differences in shape  by digitizing semilandmarks on curves and surfaces (in addition to landmarks). Those articles attached by Jonathan P. Miller provide useful information about it, and here you have another one.
