I've been using the media of STEMdiff astrocyte differentiation and STEMdiff astrocyte maturation kit for hiPSCs-derived astrocytes. Since, this media is serum free media and it claims that astrocytes after 56 days are positive for GFAP (86%). However, When I tried to see the expression of GFAP, S100 β and EAAT1 at Day 50 (mature astrocytes according to the StemCell's medium recommendations). I didn't get a good result for EAAT1 and S100 β. I am sharing the images of ICC, at Day 50 and scale bar: 100um. Please also comment about the morphology.

What can you conclude from these.
Kanza Saleem , I illustrate here astrocytes as interacting with the associated capillary. Understanding the function and "health" of the astrocyte is so very important. I see many studies that as they work to capture neuropathology data the astrocyte is missed or destroyed in the process.
