I want to test the synergistic effects between my compound and Cisplatin/Bromopyruvate in A549 non-small lung cancer cells.
I tried dissolving Cisplatin/Bromopyruvate in DMSO but the results are not encouraging. I believe for some reason DMSO interacts with these and reduces there anticancer efficacy.
Please can someone suggest proper dissolving media for Cisplatin/Bromopyruvate and the appropriate concentrations for testing their synergistic effects.
Well..Then what is the maximum stock I can prepare. I am not sure how much qunatity of drugs dissolve in how much plain H20.

I highly suggest to obtain already dissolved cisplatine in vial, ex. from oncological treatment facility cause that will not spread around so much when weighing.
How was bromopyruvate then?
Also I have the cispLatin as a powder so I really would not want to buy a new vial. Can you please suggest some solvent.
I did try 0.9% Nacl. Cisplaying did not dissolve completely. I stored it at 4oC and it still did not dissolve after overnight.

As I read cisplatin is sold to hospitals as 1mg/ ml stock solution in 0,9% NaCl with pH adjusted with NaOH and HCl, IMHO pH adjusting is very impractical and everything will be contaminated. Still I higly suggest to obtain commercial/hospital, sterile stock, it is very toxic and mutagenic, to dangerous to play with.
for cisplatin I went to oncology department and they gave me a vial with unused portion, that prevented a HUGE mess... For bromopyruvate ... pubmed has interesting option for You. Take care, all the best

as I read a fresh solution of bromopyruvate is needed each experiment ! cause it rapidly decomposes - minutes/hour
