Hello,

I would like to differentiate human naive T cells, obtained from a Buffy Coat, to Tregs cells in vitro . The protocols that I have seen involve the stimulation of naive T cells with anti-with CD3/CD28, 100 U/ml of IL2 and TGFb1 for 6 days.

I have doubts about:

1- Is it necessary to add IL2 and/or TGFb1 at day 2-3?

2- Is it necessary feeders irradiated?
