Hi, I want to grow some marine Synechococcus strains. There are many types of media used in the literature for this species, nevertheless, the one I was recommended to use (ASW = artificial seawater) is listed below. The recipe for trace metals stock solution says I should use EDTA(Na2Mg).2H2O. However, I only have Na2EDTA.2H2O available (contains no Mg). What are the possible consequences if I use the latter instead of the former? Is it going to have a dramatic influence on the availability of some metals for the alga or is it not a big deal?
Anna Segečová Are you adding the MgSO4.7H2O (which is basically just epsom salt) to your media solution aswell? Cause it will provide plenty of Mg. And I think you can get away with using the Na2EDTA.2H2O instead of the EDTA(Na2Mg).2H2O cause it only delivers a very small amount of Mg.
Laurent Lanckman yes, according to the table provided the media contains 14.2 mM MgSO4.7H2O. I am not worried about altering the amount of nutrients because the other constituents of the media provide enough Na and Mg. Nevertheless, I thought the EDTA is there to provide metals in a more bioavailable form (especially under higher pH), so I thought there must be a reason why the recipe calls for EDTA with Na AND Mg instead of just EDTA or EDTA(Na2).
Hello Anna, can I ask you which medium is this one? it's actually very simple to prepare and I am looking for a good seawater media for bacterial growth. Do you have the citation?
