I have recently started looking into repetitive PCR as a method to identify strains. I haven't done any yet myself, but I have looked at some results from other people. Something I noticed is that for one of the bacteria the result was very different (only about 60% similarity between the two runs) even though it was supposed to be the same strain, just cultured at different times. Both experiments were done using the same DiversiLab fingerprinting kit. What could be the reason for this, if contamination of the cultures is excluded? Feels like too big a variation for a normal run-to-run difference. How reliable are DiversiLab kits? I have seen some articles that say it's good, but not that many.
Hi Shukho Kim, thank you for your answer. So you are saying that if using diversilab kit which is standardised, the results should be interlaboratory comparable? And if it is done by using the standard kit and method, what is the reason that is still a big difference? What about if just one lab does the test and gets inconsistent results?
My first thought was that something was wrong with the isolated DNA but since I am not at all familiar with diversilab I was wondering if perhaps others had that issue with their kit. Thank you!
