I am preparing S30 buffers for use in making an S30 extract from E. coli BL21 (DE3) cells, but I have seen several different recipes for S30 buffers. The S30A buffer recipe I intend to use mixes 10 mM Tris-HCl (pH 8.0), 14 mM magnesium acetate, 60 mM potassium glutamate, and 2 mM DTT . However, I have seen recipes that use 14 mM magnesium glutamate instead of magnesium acetate. Is this recipe okay to use and what difference (if any) would there be in my lysate between using magnesium acetate instead of magnesium glutamate?
