I am injecting mice with Diphtheria Toxin to my mice. All the injected display neurological impairment. Did somebody experience that? Is it an effect given by the diphtheria or is the effect of the depletion I'm achieving?
I have done many Diphtheria Toxin injections in mice. We had initially quite big necrotic damage in the treated brains, we were doing intracranial injections, but eventually with a bit of fine tuning we managed to improve our protocol and achieve selective elimination of the neurons carrying the receptor.
I don't know, we had to be a lot more careful with the surgical procedure to avoid unspecific damage and adjust the dose. Maybe you can try brain injection, it may have a better outcome.
Our initial idea was to use IP delivery but it didn't work as expected.
