Hello,

Does anyone have access or can provide links to freeware that can calculate the net charge and isoelectric points (@ pH 7.4) of a peptide which contains a non-natural amino acid (ie: does not have a predefined single or triple letter amino acid code)?

AB40 natural sequence: DAEFRHDSGYEVHHQKLVFFAEDVGSNKGAIIGLMVGGVV

The sequence I am trying to calculate is a mutated version of beta-amyloid (1-40), with aspartic acid in position 7 replaced with non-natural amino acid Diaminobutyric acid.

Sequence:

DAEFRH-Diaminobutyric acid-SGYEVHHQKLVFFAEDVGSNKGAIIGLMVGGVV

Furthermore, I have a mutant peptide with glutamic acid in position 11 replaced with non-natural amino acid Ornithine.

Sequence:

DAEFRHDSGY-Ornithine-VHHQKLVFFAEDVGSNKGAIIGLMVGGVV

Does this limit me to manual calculation or the use of models? Any help will be greatly appreciated.
