Nile red staining procedure is very much straight forward. We use it to stain Lipid Droplets in zebrafish embryos. It works for both live/fixed cells. Prior to staining the cells are washed with PBS 2-3 times. For cells, 300nM of Nile red is sufficient. The cells are incubated with nile red solution for 10-15 min. The cells are then washed with PBS to remove excess dye. This is followed by imaging under green excitation light.
