When I coat cysteamine (as crosslinker for biomolecule adsorption) on gold nanoparticle treated PMMA substrate, Gold nanoparticles are desorbed. Could anyone suggest any crosslinker which will react with gold nanoparticles without affecting its monolayer formation?
How you prepared AuNP/PMMA substrate - in situ reduction method.
Did you observed any pH change while treating with cystamine?
What kind of surface coverage you have before treating with cystamine?
Glutaraldehyde won't react with AuNP. It is able to cross-link amine functionality.
Did you modified PMMA surface by any means for AuNP attachment.
PMMA was used for construction of pH responsive system. for e.g.

If the Gold nanoparticles desorbed when you added cysteamine as crosslinker, it's possible that cysteamine has a stronger interactions with AuNPs than that between PMMA and AuNPs.Thiol Group has a strong interactions with AuNPs. And the second point is maybe you added too much cysteamine.
So that, first you can try to decrease the amount of cysteamine. Second, you can try another crosslinker which has weeker interactions with AuNPs, such as with Groups of NH2, COONa, quaternary and so on.
Could you give more detail of how the AuNP is covalently attached? Thiol bonds from the PMMA?
