Hello! I need to perform a colony formation assay and to identify as viable cells, I will have to color them with MTT. Do I have to just find the MTT on the colonies?

I think that you should add the MTT solution (0m5mg/ml) per well, then incubate the cells (1-3h). Then remove the liquid and dry them. Do not use formazan solvent (DMSO/isopropanol). Just count the colonies. But the best way is to stain the colonies with crystal violet. It is cheap, quick and easy and stable for long time.

Will you use cells in suspension or for adherent cells?
