I am working on the extraction of an extremely polar compound. Many journals have suggested use of cellulose powder for column chromatography using 1-butanol saturated with water as eluent. The concentration of my product to be extracted from natural sources is about 2mg per 1000mg. I have prepared a slurry of cellulose using n-butanol saturated with water but there is no dribbling even after applying reasonable pressure. Previously, I have used silica gel for plenty of purifications and I was anticipating that cellulose will be flashing like silica gel but alas.

Can someone suggest me a method for using cellulose (sigma product code 22183-1KG-F, CAS # 9004-34-6) for the extraction of a zwitterionic product either from water (after aqueous work up) or from methanol?
