I was wondering if there are any limitations of growing cells or proliferation in collagen gels placed in 96 well plates. Like cell concentration, etc...
It depends on 1. cell culture, 2. cross-linkers in collagen gel, 3. medium and suplements and how often you will change it. Majority of cells will complete growth activity when they cover whole surface of the gel or if they are cultivated in collagen gel volume the next limitation will be expressed in oxygen concentration (poropus/nonporous gel).
Thanks for the feedback. I'm basically using immortalized cells seeded with 1M/ml concentration and renewing cell media everyday for 7 days. The cell lines I'm using are MDA-MB-231, MDA-IBC-3 and HEPG2/C3A. The matrix material is Collagen Type I extracted from rat tail.
