Qiagen DNeasy/Qiaprep spin column protocols call for either a centrifuge or vacuum to pull the tissue digest through the spin column membrane. In a pinch, could a syringe be used to create the vacuum? I'll try this in a week or two, but curious if anyone has already tried it.
While it's technically possible to use a syringe to create a vacuum, it's not recommended as it may not be as efficient or consistent as using a centrifuge or vacuum. The pressure generated by a syringe may not be enough to completely pass the tissue digest through the spin column membrane, leading to lower yields and lower purity of the extracted DNA. Additionally, there's a risk of damaging the spin column membrane with the force generated by the syringe.
