I have a compound that contains Chitosan, TGA(thioglycolic acid), SPDP (Succinimidyl 3-(2-Pyridyldithio)Propionate) , I am trying to investigate disulfide bond(between TGA and SPDP) with the help of NMR test, I am hoping if you can help me with a proper solvent, and do you think that the solvent can affect disulfide bond?
D2O might work. I doubt it will affect the disulfide bond but I can't guarantee.
SPDP is not water-soluble, so you might have difficulty with pure D2O. I would dissolve in DMSO-d6, then add as much D2O as the amount of SPDP permits. This is routine for SPDP couplings, so there should be no undesired effects on the disulfide bond.
Agree with James. Only water may cause solubility problem with SPDP& Chitosan . The other suggestions, you can also use Acetone -d6
Thanks so much for your helps, as James mentioned and I tried pure D2O couldn't solve the chitosan with SPDP, i even tried adding trifluoroacetic acid because I thought making acidic environment can help solving the sample. I will try the method that James suggested and inform you with results. Meanwhile I am open to other suggestions.
