Hi i am trying to create a bacterial transformation onto an agar plate however i do not have any miniprep DNA.
I have an old plate of the same strain that i am trying to get that is two months old that has been stored at 4 degrees but still has some colonies on it.
My question is can i use the old colonies on the old plate to create a small scale expression then purify it by miniprep and then set up a new transformation on a new plate. Or would this not work.

according to me bacterial plates older than one month old shouldn't be used ... yours is two months old! you can try inoculating in 1 ml media along with antibiotic.. you can try your luck!
It will be better if you reculture from your bacteria stock and use those colonies for your miniprep, if you do not have any stock, it will be better to reculture those old colonies onto new selection plates before you use it for miniprep
I would do the same. Try to reculture the colonies and prepare a new stock with glycerol. Ar the same time I would try to check that the new stock is the bacteria that you want doing PCR and sending for sequencing a target of 16S
