Hello,

I am performing polymerase chain reaction on lichen samples in order to sequence the ITS and beta-actin regions. Previously, my samples were working and I had high success with amplification and sequencing, but then a bout of high humidity and temperatures and extreme stormy weather has occurred over the last week and half or so and now my PCRs do not work (even though I am using the same PCR reagents and thermal cycler conditions as the times it did work).

I am wondering if anyone else has experienced poor PCR results during times of weird/extreme weather conditions (high temps, high humidity, etc.) and if you had any advice?

Thank you!
